I have been wondering this question for a very long time. Once a blade has been through tempered, is it still possible for it to go through differential hardening? Or has it already stop at through hardening?

once it's hardened and tempered, it is done. I think you are confusing your terms, hardeneing AND tempering are both prcesses in heat treating, first you harden, either the whole blade or differentially with the clay, then it is tempered. At most you could torch soften the spine, but that would not give any hamon.

It is always possible to re-heat treat a sword though. But of course, that would involve completely removing all temper that the sword has, and then repeating the entire heat treatment process using clay-coat.
The first heat treatment of a blade is always the best. Better to clay-quench initially.
